Product Description
The i-Cult-650A Culture Bag is a cell culture vessel made of specialized gas-permeable material, capable of supporting high-density growth of suspension cells.
The bag comprises two PVC tubing lines connected via needle-free injection ports – one short and one long.
The short tube ends with a rubber stopper suitable for syringe needle insertion, covered by a rubber cap to facilitate aseptic sampling or addition of culture supplements while maintaining sterility.
The long tube is equipped with a Luer connector for convenient medium replenishment. Its diameter complies with requirements for sterile tubing welders, and it can also be heat-sealed to further minimize the risk of culture contamination.
Product Materials
The bag body is constructed with gas-permeable EVA membrane, which effectively facilitates gas exchange for the cell culture medium, supporting rapid cell proliferation.
Simultaneously, it isolates the culture bag from surface and environmental contaminants such as water, other liquids, and particles, ensuring a sterile and safe cultivation process.

Usage Instructions
1、Place the culture bag holder on the work surface, ensuring the surface is clean and free of sharp objects.
2、Take a 50 mL syringe, remove the plunger, and insert the syringe barrel into the circular opening on the culture bag holder.
3、Unscrew the cap from Port A of the culture bag and connect it to the syringe barrel.
4、Add culture medium.
5、Remove air from the bag.
6、Tighten the cap on Port A.
7、Place the culture bag into the incubator.
8、When sampling for cell counting or adding cytokines is required, remove the rotary cap from Port B and use a syringe for liquid addition or extraction.
Note: The Port B cap should not be unscrewed by turning counterclockwise; instead, tighten it slightly forward (clockwise) and then pull it off directly.
9、When harvesting cells or retrieving culture medium (Note: Port A must be thoroughly disinfected before replenishing or extracting liquid).
Precautions
1、The culture bag is made of highly flexible material with strong tensile resistance.
However, care should be taken to avoid mechanical scratches during use.
Before use, please ensure that all surfaces potentially contacting the bag – such as the workbench, CO₂ incubator tray, and microscope stage – are smooth and free of sharp protrusions.
2、When adding and mixing liquids on the work surface, please avoid Zone C.
3、When moving the culture bag into or out of the incubator, hold both ends of the bag and lift it vertically from the tray surface. Avoid dragging the bag horizontally, with particular attention to Zone C.
4、Due to the relatively wide tubing diameter, to prevent liquid leakage after filling the culture bag with medium, please ensure the tubing end is tightly closed using a clamp, secured with a knot, and then placed in the incubator,
as illustrated below:
